The Third Institute of Physics at the Georg-August-Universität Göttingen is seeking to appoint a
Ph.D. Research Associate
This role is part-time, representing 65% of a full-time position, with an initial contract duration of three years. The compensation will be aligned with Pay grade 13 TV-L.
Key ResponsibilitiesThe viscoelastic characteristics of cellular structures are crucial in various biological activities that involve significant morphological changes, such as adhesion, spreading, growth, contraction, division, migration, tumorigenesis, and tissue formation. It is generally acknowledged that the primary reaction to external forces stems from the actin cortex, which is directly linked to the plasma membrane. This membrane/cortex composite acts as a dynamic boundary that governs the cell's shape and undergoes remodeling over seconds due to protein turnover and myosin-driven contractions, thus enabling cells to swiftly adapt and respond to external stimuli. Our objective is to differentiate between intrinsically passive and active properties of the cortex shell. We aim to evaluate the hypothesis that passive recruitment of surplus surface area, poroelastic relaxation of the cytosol, and active contraction of the cortex represent cellular responses occurring over distinct time frames. The successful candidate will incorporate a micropipette aspiration system into an atomic force microscope (AFM) that is connected to a spinning disk fluorescence microscope. This setup will facilitate the investigation of the plasma membrane reservoir's role and the membrane/actin cortex interaction in epithelial cells and cardiomyocytes as they respond to controlled stretching of the cellular surface.
This position is designed to nurture emerging researchers and enable the successful candidate to pursue a doctoral degree.
QualificationsIdeal candidates should possess a Master's degree in Physics, Biology, or Chemistry (or an equivalent qualification) with relevant experience in the biophysics of cellular mechanics utilizing biological model systems. Expertise in cellular biophysics, advanced light microscopy, and atomic force microscopy/colloidal probe microscopy is highly desirable. The applicant should demonstrate curiosity, high motivation, self-organization, and a strong interest in collaborating within an interdisciplinary team to conduct pioneering research.

Every biological system utilizes fundamental principles of physics to carry out its daily functions. Our research focuses on understanding how mechanical properties are involved and sometimes even dictate the correct and robust interactions among various biological components. While cell and tissue mechanics have been a focal point of research over the past two decades, we are only beginning to explore the complex interdependencies between biochemical signaling, mechanical forces, and viscoelastic properties. The quantitative characterization and mathematical modeling of these intricate interactions within cells and tissues are central to our laboratory's mission. We are developing innovative measurement techniques to quantify forces and tensions in three-dimensional tissues, membranes, and filaments. Additionally, we are exploring new microscopy and analytical methods, integrating them with quantitative assessments of viscoelastic properties in cells and tissues.
