Position Title: Externally Funded Doctoral Student (m/f/d) - Epigenetic Regulation of Pancreatic Cancer Cell Plasticity

Company Overview:

The research team led by Prof. Steven Johnsen operates within the Robert Bosch Center for Tumor Diseases (RBCT). This newly established non-profit translational cancer research institute is located at the Bosch Health Campus, adjacent to a prominent hospital. As part of the National Center for Tumor Diseases (NCT) Southwest and the Comprehensive Cancer Center Tübingen-Stuttgart, the RBCT is committed to pioneering cancer research and developing innovative treatment methodologies.
The Johnsen group specializes in deciphering intricate gene regulatory networks that influence pancreatic cancer development. We employ cutting-edge techniques, including various genome and transcriptome sequencing methods, high-throughput CRISPR/Cas9 screening, multiplex immunofluorescence staining, single-cell RNA sequencing, and genetic engineering on tumor cell lines, primary patient samples, and translational tumor models. Our primary research objective is to uncover gene regulatory mechanisms that can serve as novel therapeutic targets to enhance treatment efficacy and improve survival rates for pancreatic cancer patients.

Job Responsibilities:

- Employ modern sequencing-based molecular biology techniques (such as RNA-seq, ChIP-seq, HiChIP, Hi-C, PRO-seq) to test hypotheses and investigate epigenetic and gene regulatory mechanisms governing pancreatic tumor cell identity and therapeutic responses.
- Conduct bioinformatic analysis and interpretation of data, followed by hypothesis testing and formulation.
- Execute multiplex immunofluorescence staining to validate gene regulatory mechanisms in primary tumor tissues using advanced multispectral imaging technology.
- Prepare histological samples (FFPE tumor material) for single-cell RNA sequencing utilizing 10X Genomics technology.
- Carry out sophisticated cell culture experiments to assess the effects of drug treatments across various translational model systems.
- Engage in both internal and external research collaborations.
- Document work processes, data collection, and analyses.

Qualifications:

- A Master of Science degree in a relevant field (biology, molecular biology, biotechnology, or similar) is required.
- Candidates must have secured or be capable of securing external funding for their position (e.g., doctoral fellowship from recognized funding bodies).
- Experience in cell culture, transfections, and related techniques is essential.
- Extensive background in molecular biology techniques (real-time PCR, Western blotting, etc.) is necessary.
- A strong interest in acquiring proficiency in bioinformatics analysis is required.
- Candidates should demonstrate a robust work ethic, high self-motivation, and the ability to work independently as well as collaboratively.
- Proficient written and spoken English skills are mandatory.
