Project Overview:
This is a fixed-term research project focused on understanding the formation of a functional liver during development. The research conducted by the Ober group aims to elucidate the cellular and molecular mechanisms that coordinate the intricate assembly of the complex three-dimensional liver architecture during both development and regeneration. Our primary focus is on the cellular behaviors and interactions that drive the differentiation of hepatic progenitors into the specialized organization of the mature organ, utilizing zebrafish as a model organism.
During liver development, bipotent progenitors differentiate into hepatocytes and biliary epithelial cells, contributing to the formation of the biliary ductal network, while endothelial cells concurrently establish the blood vessel network. Recent findings have highlighted the critical roles of endothelial cells in the differentiation of both functional hepatocytes and the biliary network. Comprehensive proteomic analyses have identified several novel candidate factors, including cytoskeletal regulators and extracellular matrix components. This PhD project will explore the molecular and cellular roles of endothelial cells in establishing a functional liver architecture during development, with a particular emphasis on the polarization and connectivity of hepatocytes to the biliary ductal network.
Research Techniques:
The successful candidate will employ zebrafish genetics alongside cutting-edge molecular biology techniques, including CRISPR/Cas9 genome editing and transgenesis. Additional methodologies will encompass fluorescent in situ hybridization, immunohistochemistry, and both live and fixed-tissue imaging techniques, combined with quantitative image analysis and statistical evaluation.
Training and Development:
The selected PhD student will receive comprehensive training in molecular biology, zebrafish husbandry and genetics, fluorescence microscopy, and computational image analysis.
Research Environment:
The Ober group is part of a vibrant and collaborative team within the Division of Developmental Biology at Friedrich-Alexander University (FAU) Erlangen-Nürnberg, Germany. FAU and its affiliated institutes offer an exceptional research environment for developmental biology, cell biology, and regeneration, equipped with all necessary facilities to support this ambitious research initiative.
Compensation:
Salary will be aligned with TV-L E13, at 50-65% based on experience, for a full-time commitment over a duration of three years.
