Join our dynamic research group dedicated to advancing the field of neurooncology as a PhD student. The Translational Neurooncology Junior Group, funded by the Mildred Scheel Early Career Center (MSNZ) and the Frankfurt Cancer Institute (FCI) is under the supervision of Dr. Ann-Christin Hau and Dr. Tanja Müller. Affiliated with the Dr. Senckenberg Institute of Neurooncology and the Neurological Institute (Edinger Institute) of the Goethe University Hospital Frankfurt am Main, our team is at the forefront of pre-clinical research. Our group contributes to the evolvement of patient-derived models of primary brain tumors, deploying them for genetic (CRIPR-Cas) and pharmaceutical screens through live cell imaging. Our commitment lies in unravelling the molecular mechanisms of tumor evolution and tumor invasion, with the ultimate goal of advancing understanding and treatment of these complex diseases.
Research Topic: Join us as we confront glioblastoma, a challenging primary brain tumor known for its high recurrence rates and grim prognosis. Our advertised PhD project focuses on developing a pioneering pre-clinical ex vivo glioblastoma organoid model using relapsed tumor samples. We integrate patient-specific tissue, immune cells, and genetically engineered HER2-specific CAR-NK cells to explore innovative treatment avenues. Traditional therapies struggle against glioblastoma's immunosuppressive microenvironment. Our organoid model aims to inspire the development of novel immunotherapeutic strategies. To bolster the reliability of our findings, we'll validate our research using immunocompetent mouse models, particularly humanized mice. Additionally, we have the unique opportunity to contribute to the ongoing CAR2BRAIN study, the first-in-human clinical trial using NK cells. This ensures our work translates effectively to clinical settings, potentially improving outcomes for patients. Join us and be a part of meaningful change

Your Role
We are seeking a motivated and dedicated PhD student to:

- Generate glioblastoma organoids from relapsed tumor samples,
- Optimize organoid culture protocols to ensure robust and standardized organoid formation,
- Co-culture patient-derived organoids with autologous immune cells and genetically engineered CAR-NK cells targeting specific tumor antigens,
- Evaluate treatment efficacy and responses in HER2-positive glioblastoma patients using established organoid model.
- Generation of personalized and humanized mice: intracranial injection of patient-derived organoids into mouse xenografts along with human immune system reconstitution.

Who you are
- Hold - or will obtain before start of the position - a master degree in immune biologie, biomedicine or a related field.
- Demonstrate exceptional academic achievements throughout your Bachelor's and Master's studies.
- Possess relevant expertise in organoid culture techniques, immune cell biology and animal experiments (FELASA B)
- Are fluent in academic English (speaking and writing). Proficiency in German advantageous but not mandatory.
- Exhibit strong motivation to pursue a doctoral degree with this impactful project.
- Proven ability to work independently and collaboratively in research environment.
- Effective Communication and presentation skills.
- Strong problem-solving skills and a proactive approach to research challenges.
- Previous experience in cancer research, particularly with a focus on glioblastoma and immunotherapy. Due to legal regulations, a valid proof of measles immunity / measles vaccination required.
