The Doctoral candidate (f/m/d) Operational area: Clinic for Gastroenterology, Metabolic Diseases and Internal Intensive Care Medicine -AG Strnad- (Med. Clinic III)  Working hours: Part-time 65% (currently 25,3 hours/week)  Duration/Term of employment: initially limited to 3 years in accordance with the WissZeitVG. The successful candidate is expected to pursue a doctoral degree. Start date: as soon as possible Remuneration/Rates: EG 13 (TV-L) Project:  Alpha-1 antitrypsin (AAT) is a secreted hepatocellular protein. AAT mutations cause AAT retention in the endoplasmic reticulum (ER), leading to AAT deficiency (AATD). Liver disease is one of the leading causes of mortality in AATD, but the hepatic manifestation displays great interindividual variability. AATD is an established model for proteotoxic stress. Over the past 10 years, our clinic has put together a globally unique cohort of AATD patients with associated biomaterials. During your doctoral thesis, you will take advantage of these samples to investigate the pathogenesis of AATD and the impact of various factors on disease development. In vivo conditions will be examined with patient-derived induced pluripotent stem cells (iPSCs), and human samples will be examined using deep visual proteomics (DVP) and single-cell proteomics (SCP). A special focus will be the evaluation of spatial transcriptomic analyses of AATD livers. Your tasks: Bioinformatic analysis of various omics measurements (SCP, spatial transcriptomics, RNAseq) Validation of the identified signaling pathways in cell/model organelles (cell culture, iPS Biochemical, molecular biological, and pathological analyses Your profile: Successfully completed university degree (diploma, master's or equivalent) in biology, biotechnology, biochemistry, pharmacy, or human biology/molecular medicine Experience in experimental/bioinformatic research and interest in immunology, cell and molecular biology Desired expertise: Good written and spoken English skills (level C1), basic German knowledge Basic knowledge of bioinformatics Experience  in bioinformatic data analysis in experimental research and interest in bioinformatics, immunology, cell and molecular biology  with transgenic animal models and model organisms with essential techniques of cell and molecular biology in cell culture Ability to work in a team, flexibility, and structured working style Enjoyment of cooperative and interdisciplinary work Strong interest in new methods Interest in basic and translational research Why choose us?
