The research will focus on the hexanucleotide repeat expansions in the C9ORF72 gene, recognized as the leading genetic factor in amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). The pathological mechanisms associated with C9ORF72-related ALS/FTD involve both loss and toxic gain of functions. Despite growing evidence regarding loss of function mechanisms, the cellular and physiological roles of C9ORF72 and their significance in ALS pathogenesis remain inadequately explored. Our analysis of C9ORF72 proteomics in motoneurons, along with co-immunoprecipitation experiments, has indicated a notable interaction between C9ORF72 and ribosomes. These interactions are crucial for the translation elongation phase, as targeting C9ORF72 through shRNA significantly diminished translation capacity in cultured motoneurons, particularly in axonal regions. This research initiative aims to elucidate how C9ORF72's interaction with ribosomes influences translation in motor axons and to characterize the pathological implications of this interaction on axonal and synaptic degeneration in ALS.
The prospective PhD candidate will investigate the role of C9ORF72 in regulating translation elongation in both wildtype mouse and human motoneurons, focusing on axonal and presynaptic compartments. The candidate will also explore whether the loss of this function correlates with pathomechanisms that lead to reduced translation capacity in axons in ALS models. The project will involve working with primary motoneurons from mice and human-induced pluripotent stem cell (hiPSC)-derived motoneurons from ALS patients with C9ORF72 repeat expansions, utilizing super-resolution microscopy and biochemical assays. These investigations will provide valuable insights into the loss of function mechanisms relevant to ALS axonopathy and neurodegeneration.
