We are the UKE - the University Medical Centre Hamburg-Eppendorf [Universitätsklinikum Hamburg-Eppendorf]. A pulsating centre of health in the heart of Hamburg. The motivated research group Vascular Biology studies the impact of matrix stiffness changes on endothelial cell behavior in vascular morphogenesis and disease. This project aims to characterize novel stiffness biosensors in endothelial cells in vitro and in vivo. Our research group employs a variety of transgenic mouse models, primary cell culture systems and state-of-the-art microscopy methods such as live imaging, super-resolution and nanoindentation microscopy.

Main techniques used in this project:

- Generation and characterization of transgenic mice
- Molecular cloning
- Isolation and culture of primary endothelial cells and lentiviral gene expression in endothelial cells
- Hydrogel techniques for 2D and 3D cell culture
- qPCR analysis and Western blot analysis
- Whole mount immunofluorescence staining and confocal microscopy
- Quantification of imaging data, quality control, large datasets and statistical analysis
- Planning of experiments, preparation and participation in scientific publications and congresses

This position is to be filled within the framework of a third-party funded project for a limited period of one year with a possibility of extension and can also be filled on a part-time basis.

- A strong track record with peer-reviewed publications
- In-depth interest in vascular biology, endothelial cell biology and mechanobiology
- Desirable are experiences in endothelial cell biology, developmental biology or mechanobiology, experimental animal work (FELASA course), and applied laboratory methods: primary cell culture, 2D matrix gel preparation, qPCR, advanced fluorescence microscopy and atomic force microscopy.
- Ideally Adobe Illustrator, ImageJ, and GraphPad Prism knowledge
- Ability to work independently, organizational skills, teamwork, quick comprehension, commitment
