The Institute of Pharmacology and Toxicology aims to decipher disease mechanisms to restore the function of diseased organs, particularly the heart. Utilizing various in vitro and in vivo models, we investigate pathophysiological molecular mechanisms and signaling pathways underlying cardiovascular diseases, thereby developing new targets for clinical translation. The primary focus of the "Immunopharmacology of the Heart" research group (headed by Dr. rer. medic. Erik Klapproth) at the Institute of Pharmacology and Toxicology, TU Dresden, lies in examining the role and interaction of different immune cell subpopulations following an acute myocardial infarction. Specifically, the Klapproth group focusses on the significance of neutrophils for post-infarction pathogenesis and remodeling, as well as the chemokines and proteases crucial for immune cell recruitment.
Starting at the next possible time, we are looking for a
PhD Candidate (f/m/x) in Immunopharmacology of the Heart
in a part time position. This position is initially limited to 36 months, with remuneration according to the Collective Agreement for the Public Service of the States (TV-L), entailing possible placement in pay group E13 TV-L contingent upon personal qualifications.
Aufgaben
The doctoral project aims to elucidate the epigenetic and metabolic shifts in hematopoietic stem and progenitor cells post-heart attack, favoring a more aggressive, inflammatory phenotype of myeloid cells (myeloid bias). Additionally, we seek to understand the role of this phenotypic switch in the occurrence of secondary cardiovascular events and the development of cancer subsequent to the primary infarction, with a particular focus on the significance of A disintegrin and metalloprotease 10 (ADAM10) for these processes. In previous studies, our research group has already highlighted the fundamental role of ADAM10 in the regulation of neutrophil infiltration, survival, and heart function post-myocardial infarction, providing the foundation for this PhD project. To address these research questions, we utilize mouse models with cardiac-specific ADAM10 knockout, specific pharmacological inhibitors, as well as various cell culture models. To characterize ADAM10 function under physiological and pathophysiological conditions, we combine numerous in vivo techniques with OMICS approaches such as mass spectrometry or bulk and single-cell RNA sequencing, as well as flow cytometry (FACS), histology and immunofluorescence microscopy.
Profil
- completed degree in Biology, Biochemistry, Biophysics, or a related natural science field
- high motivation and teamwork skills, pleasure in independent work and practical problem-solving strategies in a scientific environment
- interest and willingness in working with innovative transgenic animal models (prior experience with mice handling and possession of FELASA-B certification are advantageous)
- practical experience in molecular biology (such as Western blot, qPCR, cell culture, immunofluorescence)
- high level of personal commitment
